Application: Pharmacogenomic and molecular epidemiological study of HIV/AIDS patients co-infected with Hepatitis C virus (HCV)

Objectives: The objectives of this project are:
  1. specific primers, probes and real-time PCR assay will be designed and optimized to detect the SNPs for IFNL4 among HIV/HCV co-infected patients;
  2. treatment history and clinical responses from individual patients will be retrieved from Integrated Treatment Centre. Statistical analyses will be carried out to evaluate the impacts of IFNL4 SNPs on peg-IFN/RBV responsiveness;
  3. the prevalence of IFNL4 TT/rG, IL-28B rs12979860 T/C and rs8099917 G/T polymorphisms will be estimated among 80 retrospective and 30 prospective HIV/HCV co-infected patients. Statistical analyses will be carried out to evaluate the relationship between these 3 SNPs as well as the contribution of confounders; and
  4. partial HIV-1 pol gene and HCV NS5B sequence regions of 110 samples will be amplified and analyzed by bioinformatics software to infer the phylogenetic relationship.
